Hypoxyprobe公司基于缺氧探針抗體技術，在此技術上開發(fā)了系列缺氧探針標記試劑盒，為組織疾病的診斷和治療提供有力研究工具。
檢測原理
低氧標志物測定是利用還原硝基有選擇性結合乏氧細胞的能力而形成的一種從細胞水平測定腫瘤乏氧的技術，其原理是：腫瘤乏氧細胞還原能力強，當具有電子親合力的硝基咪唑主動擴散透過細胞脂膜，在細胞內(nèi)硝基還原酶作用下，硝基被還原，還原產(chǎn)物與大分子物質(zhì)不可逆結合，滯留在組織內(nèi)。而正常含氧細胞親電子硝基咪唑還原產(chǎn)物立即被氧化，從而用硝基咪唑類化合物在腫瘤內(nèi)的代謝程度來反映腫瘤的乏氧
Pimonidazole是新型的乏氧細胞標記物，HPI公司擁有硝基咪唑類化合物特異性抗體，該抗體為公司技術，可用于免疫組織化學、酶聯(lián)免疫吸附試驗和流式細胞分析檢測。
Pimonidazole 應用
特異性的低氧探（Hypoxyprobe-1）zui初用于反映腫瘤細胞內(nèi)的低氧梯度，正常細胞無表達或低表達。而在低氧的腫瘤細胞內(nèi)可見高表達，并且隨低氧的程度越重而顏色越深。由于其很好的化學穩(wěn)定性、水溶性，同時小分子的探針可以自由穿過血腦屏障而到達腦組織細胞及腦腫瘤細胞。在實驗中應用低氧探針可以形象直觀的通過免疫組化來分析腦挫傷周圍組織細胞低氧的范圍和程度，同時對實驗設備及器材要求較低，為無創(chuàng)性實驗。
